Moderate human evidenceEndogenous circadian hormone / supplement

Melatonin

Melatonin is a hormone involved in night-time signaling and circadian timing. Supplement effects depend on the sleep problem and context; it is not a universal sleep treatment.

Also indexed as: N-acetyl-5-methoxytryptamine

A clear starting point

Melatonin helps signal biological night. That is different from switching the brain off. Human sleep trials show modest average effects, and results vary between people and sleep conditions.

Systems mapping shows biological relevance, not a treatment recommendation or a patient CPI score.

What does the evidence show?

Small average sleep improvements in the cited meta-analysis. Circadian timing and insomnia are different clinical questions.

What this does not establish

A pooled average does not predict individual response. Long-term and pediatric questions require dedicated assessment.

Meta-analysis: melatonin for the treatment of primary sleep disorders.

PloS one · 2013 · Journal Article; Meta-Analysis; Research Support, N.I.H., Extramural; Research Support, Non-U.S. Gov't · PMID 23691095

Verified sample: 1,683 · Adults and children with primary sleep disorders across 19 studies

Modest average improvements in sleep onset, total sleep time and sleep quality.

Limitations: Heterogeneous populations and formulations; not a universal insomnia recommendation.

Safety is a separate question

Drowsiness, headache and dizziness may occur. Supplement content can vary; avoid assuming natural means risk-free.

Contraindications & cautions

Pregnancy, breastfeeding and use in children warrant clinician assessment; do not self-treat unexplained persistent sleep problems.

Medication & supplement interactions

Sedating medicines may increase drowsiness. A pharmacist should review anticoagulants and other concurrent medicines.
